  I am a botanist with expertise in plant identification and authentication through the use of macro-microscopic histological characteristics observed with various microscopes such as light, fluorescence, polarizer, confocal, and scanning electron microscopes.

  It's fascinating how every plant has its own unique features. Personally, I find the Asparagus plant particularly interesting due to its distinctive characteristics and its versatility as both a food and medicine.

  My typical day involves identifying and authenticating the Plants using Microscopic characters. Mainly to recognize the critical characters for botanical authentications.

Thanks for sharing the images and report of your experiment. I appreciate the effort you all put into this research. Your report's positive note highlights the improvement that needs to be carried out. The scale bar, reference images, and comparison with the same magnification of both experimental setups are highly advisable for the proper result interpretation.

My minor suggestions are

If possible, try rearranging the photos by comparing the close view of the stomata and the single cell of both light and dark setups.

Include materials and methods used: name of the species used for the study, the number of leaves collected and from which region of the plant (above, middle, or below), and the maturity level of the leaf used. What microscope (model, make, objective and the fluorescence filter used). All these data improve your report for understanding the experiment and output of the results.

I want to suggest some points.

1. Chloroplasts move towards the light source; thus, they mostly align close to the cell wall. I guess what you see as red blobs is due to clusters. Unless I see the image, I can't confirm this.  

2. What type of microtome do you have? If you are doing alcohol-xylene treatment for microtome sections, you won't see any chloroplast; microtome sections are good for structural views, not for cell contents.  

The plane of sectioning is also important for estimating the chloroplast. The best is a surface view (depending on which leaf you take for studies). If cross-section, it is important to ensure sections are from the leaf's upper, meso, and lower portions.
